Native Chemical Ligation

Native chemical ligation is a chemoselective method for assembling peptide segments into larger proteins, enabling precise access to sequences that are difficult to produce biologically. It joins a peptide bearing a C-terminal thioester to another with an N-terminal cysteine through transthioesterification, followed by an intramolecular S-to-N acyl shift that forms a native peptide bond. In immunology and infection research, this strategy supports the synthesis of defined antigens, pathogen-derived proteins, and post-translationally modified peptides for studying immune recognition, antibody binding, and host-pathogen interactions. It also helps generate tailored protein reagents for structural and functional experiments.
