Ros Detection Assay

ROS detection assays are analytical methods used to measure reactive oxygen species, chemically reactive molecules that influence redox signaling, inflammation, and host defense. In a typical assay, a redox-sensitive probe enters cells or a sample and undergoes oxidation, producing a measurable change in fluorescence or absorbance that reflects ROS generation under defined conditions; signal intensity is compared with appropriate controls. In immunology and infection research, these assays help quantify oxidative bursts in immune cells, assess pathogen-triggered responses, and evaluate how oxidative stress affects microbial survival or tissue injury. Their results support studies of antimicrobial mechanisms and inflammatory disease.

Cut sheath segments and leaf discs are exposed to a bacterial flagella-derived peptide functioning as a PAMP. Upon recognition of the peptide via pattern recognition receptors (PRRs) on plant cells, a rapid and transient production of ROS occurs, which is detected via a chemiluminescence assay.
