Thrombin Activation

Thrombin activation is the enzymatic conversion of inactive prothrombin into thrombin, a central step linking blood coagulation with inflammation and host defense. In the prothrombinase complex, factor Xa cleaves prothrombin on phospholipid surfaces in the presence of factor Va and calcium, producing active thrombin. Thrombin then converts fibrinogen into fibrin, activates platelets and additional coagulation factors, and can signal through protease-activated receptors on immune and vascular cells. In immunology and infection, this pathway helps contain tissue injury and pathogens through clot formation, but excessive or dysregulated activation can promote inflammation, thrombosis, and tissue damage.

Purification and Activation of Platelets from Murine Whole Blood

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.

Measurement of Factor V Activity in Human Plasma Using a Microplate Coagulation Assay

This study describes a novel microplate assay that measures FV coagulation activity during fibrin clot formation in human plasma which has not been reported previously. The method uses a kinetic microplate reader to continuously measure the change in absorbance at 405nm during fibrin clot formation in human plasma.

Measurement of Larval Activity in the Drosophila Activity Monitor

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.
