Tonsil Cell Isolation

Tonsil cell isolation is a laboratory method for obtaining viable immune and epithelial cells from tonsil tissue, enabling direct study of local human immune responses. The tissue is mechanically dissociated, often with optional enzymatic digestion, and the resulting suspension is filtered and centrifuged to remove debris and enrich cells; viability depends on prompt, gentle handling and appropriate buffered conditions. Isolated tonsil cells can be characterized by flow cytometry, cultured, or exposed to pathogens and immune stimuli to examine lymphocyte activation, antigen presentation, cytokine production, and infection-related responses. This approach supports research into mucosal immunity, host-pathogen interactions, inflammation, and translational immunology.

Palatine tonsils are a rich source of B and T lymphocytes. Here we provide an easy, efficient and rapid protocol to isolate B and T lymphocytes from human palatine tonsils. The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.
