Elastin Visualization

Elastin visualization is the detection and imaging of elastin fibers, durable extracellular-matrix proteins that provide stretch and recoil to tissues such as arteries, lungs, skin, and connective tissue. In medical research, tissue sections can be examined with elastin-selective histological stains, immunolabeling, or fluorescence and electron microscopy, which generate contrast from the fibers’ chemical properties, antigenic markers, or structural organization. These approaches reveal elastin distribution, fragmentation, and remodeling in healthy and diseased tissue. Elastin visualization supports diagnosis and pathology studies of vascular, pulmonary, and connective-tissue disorders, while helping researchers evaluate tissue engineering strategies and changes associated with aging or injury.

Elastin-like polypeptides are stimulus-responsive biopolymers with applications ranging from recombinant protein purification to drug delivery. This protocol describes the purification and characterization of elastin-like polypeptides and their peptide or protein fusions from Escherichia coli using their lower critical solution temperature phase transition behavior as a simple alternative to chromatography.

Tri-layered Electrospinning to Mimic Native Arterial Architecture using Polycaprolactone, Elastin, and Collagen: A Preliminary Study

The aim of this study was to mimic the native three layered architecture of the arterial wall. To accomplish this, electrospinning was employed with the use of a 3-1 (input-output) nozzle and blends of polycaprolactone, elastin, and collagen.

Recombinant protein-engineered hydrogels are advantageous for 3D cell culture as they allow for complete tunability of the polymer backbone and therefore, the cell microenvironment. Here, we describe the process of recombinant elastin-like protein purification and its application in 3D hydrogel cell encapsulation.
