Eus-bd

EUS-BD, or endoscopic ultrasound-guided biliary drainage, is a minimally invasive technique that creates an internal pathway for bile flow when conventional endoscopic drainage is unsuccessful or not feasible. During the procedure, an echoendoscope uses ultrasound to identify an obstructed bile duct or the gallbladder, which is punctured through the gastrointestinal wall; a guidewire then supports placement of a stent to bypass the blockage and restore drainage. In medicine, EUS-BD provides an alternative to percutaneous drainage for selected patients with biliary obstruction, including those with malignant disease, and can relieve jaundice and related symptoms while expanding therapeutic options in advanced endoscopy.

Axonal transport of BDNF, a neurotrophic factor, is critical for the survival and function of several neuronal populations. Some degenerative disorders are marked by disruption of axonal structure and function. We demonstrated the techniques used to examine live trafficking of QD-BDNF in microfluidic chambers using primary neurons.

Recombinant BDNF containing an Avi sequence (BDNFAvi) is produced in HEK293 cells in a cost-effective manner and is purified by affinity chromatography. BDNFavi is then directly mono-biotinylated with the enzyme BirA in a tube. BDNFavi and mono-biotinylated BDNFavi retain their biological activity when compared to commercially available BDNF.

Culturing Murine Spiral Ganglion Neuron Explants on Multielectrode Arrays

This video demonstrates the process for isolating spiral ganglion explants from the inner ear and co-culturing them with the organ of Corti on a multielectrode array. Spiral ganglia are carefully extracted and sectioned into smaller tissue sections or explants. These explants are cultured on the multielectrode array's active area to promote neuronal process outgrowth.

The mechanism underlying the therapeutic effects of Deep Brain Stimulation (DBS) surgery needs investigation. The methods presented in this manuscript describe an experimental approach to examine the cellular events triggered by DBS by analyzing the gene expression profile of candidate genes that can facilitate neurogenesis post DBS surgery.

Spiral Ganglion Neuron Explant Culture and Electrophysiology on Multi Electrode Arrays

We present a protocol to culture primary murine spiral ganglion neuron explants on multi electrode arrays to study neuronal response profiles and optimize stimulation parameters. Such studies aim to improve the neuron-electrode interface of cochlear implants to benefit hearing in patients as well as the energy consumption of the device.
