Graft Function Assessment

Graft function assessment is the systematic evaluation of how well a transplanted organ or tissue is performing after transplantation, helping clinicians identify successful integration, dysfunction, or rejection. It combines the graft’s clinical performance with laboratory measurements, imaging, and, when necessary, tissue biopsy to detect changes in organ-specific function and structural injury. In medicine, serial assessments establish a baseline, track recovery, and distinguish expected postoperative changes from complications such as ischemia, infection, drug toxicity, or immune-mediated rejection. Timely interpretation guides immunosuppressive management and other interventions, supports long-term graft survival, and provides objective outcomes for transplant research.

Marginal grafts, such as fatty livers, grafts from older donors, or livers retrieved after cardiac death (DCD) tolerate conventional, cold static storage only poorly. We developed a novel model of subnormothermic ex vivo liver perfusion for preservation, assessment, and repair of marginal liver grafts prior to transplantation.

Capillaroscopy is a non-invasive, relatively inexpensive methodology for directly visualizing the microcirculation. The forearm blood flow technique provides accepted non-invasive measures of endothelial function.

Mouse Models for Graft Arteriosclerosis

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

The cell membrane modification of red blood cells (RBCs) with hyperbranched polyglycerol (HPG) is presented. Modified RBCs were characterized by aqueous two phase partitioning, osmotic fragility and complement mediated lysis. The camouflage of surface proteins and antigens was evaluated using the flow cytometry and Micro Typing System (MTS) blood phenotyping cards.
