Orthotopic Glioma Model

An orthotopic glioma model is an experimental system in which glioma cells or tumor tissue are implanted into the corresponding anatomical site in the brain, reproducing key features of tumor growth in its native environment. The implanted cells interact with surrounding neural tissue, blood vessels, extracellular matrix, and immune components, allowing tumors to develop within the brain microenvironment and form clinically relevant patterns of invasion. In medicine, these models support investigation of glioma biology, tumor progression, treatment response, and mechanisms of therapeutic resistance. They are particularly valuable for evaluating surgery, radiotherapy, drug delivery, and emerging targeted or immun-based therapies before clinical translation.

Two techniques can be used to establish this model: injection of a cancer cell suspension into the cecal wall or transplantation of a piece of subcutaneous tumor onto the cecum. This model is useful for studying the natural progression of colorectal cancer and testing new therapeutic agents against colorectal cancer.

Generation of an orthotopic mouse model of anaplastic thyroid carcinoma is described here. This technique employs surgical placement of human anaplastic thyroid cancer cells into the thyroid of immunodeficient mice, thus creating a more clinically relevant setting to study disease progression as well as screen innovative therapeutic interventions.
