Skin Temperature Analysis

Skin temperature analysis is the measurement and interpretation of temperature patterns at the body’s surface, providing noninvasive information about physiological changes. It typically uses contact sensors or infrared thermography to detect emitted heat, while local temperature reflects interactions among blood flow, metabolism, environmental conditions, and heat exchange with the surroundings. In medicine, clinicians and researchers use these measurements to monitor circulation, inflammation, wound healing, fever, and responses to treatment. Comparing temperature across body regions or over time can reveal clinically relevant changes, support patient monitoring, and contribute to research on thermoregulation and disease processes.

We describe a protocol to efficiently isolate skin resident T cells from human skin biopsies. This protocol yields sufficient numbers of viable human skin resident lymphocytes for flow cytometric analysis and ex vivo culture.

The skin is home to a complex immune cell network. We describe an efficient methodology for the digestion of mouse skin, from different parts of the animal's body, in order to obtain a single-cell suspension and analyze the different leukocyte populations resident in the skin by flow cytometry.
