Stair Climbing

Stair climbing is a form of weight-bearing aerobic exercise in which repeated ascent requires the body to lift its mass against gravity. The movement engages the quadriceps, hamstrings, gluteal muscles, and calf muscles while increasing heart rate, ventilation, and oxygen demand to support working tissues. In medicine, stair-climbing assessments can provide a practical indication of functional capacity, lower-limb strength, balance, and exercise tolerance. Regular stair climbing may contribute to cardiovascular fitness and metabolic health, while performance changes can help clinicians monitor mobility, recovery, and the effects of rehabilitation or chronic disease.

We present an optimized inexpensive and reliable negative geotaxis assay in Drosophila melanogaster as a model for neurodegenerative disorders. Being more sensitive to mild locomotor defects, this assay will help screen for potential genetic interactions and drug targets.

This step-by-step protocol analyzes Drosophila negative geotaxis behavior using an automated multi-cylinder system that hosts hundreds of flies and synchronizes their action by an electric motor. Upon synchronization, fly negative geotaxis behavior is assayed, digitally recorded, and analyzed using the self-designed RflyDetection software.

Here we describe two assays that have been established to study age-dependent neurodegeneration of dopaminergic (DA) neurons in Drosophila: the climbing/startle-induced negative geotaxis assay which allows to study the functional effects of DA neurons degeneration and the tyrosine hydroxylase immunostaining which is used to identify and count DA neurons in whole brain mounts.
