Transcatheter Edge-to-edge

Transcatheter edge-to-edge repair (TEER) is a minimally invasive cardiac procedure that treats valvular regurgitation by bringing valve leaflets together to improve closure. It uses a catheter, guided by echocardiographic imaging, to deliver a clip or similar device through a blood vessel and grasp the leaking leaflets, creating smaller valve openings that limit backward blood flow. In medicine, TEER can reduce symptoms and improve cardiac function in selected patients with mitral or tricuspid regurgitation, particularly when conventional surgery carries substantial risk. Its catheter-based approach provides a less invasive option for patients who may not tolerate open surgery and supports management of structural heart disease.

The full process from brain specimen preparation to serial sectioning imaging using the Knife-Edge Scanning Microscope, to data visualization and analysis is described. This technique is currently used to acquire mouse brain data, but it is applicable to other organs, other species.

In this protocol, we present the experimental procedures of a cell spreading assay that is based on live-cell microscopy. We provide an open-source computational tool for the unbiased segmentation of fluorescently labeled cells and quantitative analysis of lamellipodia dynamics during cell spreading.
