Dissociated Cell Culture

Dissociated cell culture is a laboratory method that separates cells from tissue and maintains them individually or in small clusters under controlled conditions, enabling direct study of cellular behavior. In neuroscience, tissue is typically disrupted through mechanical or enzymatic dissociation, then the resulting neurons and glial cells are plated on a suitable substrate and supported with defined culture media. This approach provides access to neuronal morphology, survival, differentiation, synapse formation, and cell-to-cell signaling in a simplified experimental system. Researchers use dissociated neural cultures to investigate development and disease mechanisms, test pharmacological treatments, and evaluate cellular responses that may be difficult to observe within intact brain tissue.

Primary dissociated midbrain dopamine cell cultures allow for the study of presynaptic characteristics of dopamine neurons. They can be used to monitor real-time dopamine release kinetics and protein/mRNA levels of regulators of dopamine exocytosis. Here, we show you how to generate these cultures from rodent neonates.
