Hindbrain Progenitors

Hindbrain progenitors are developing neural cells that generate the neurons and glia of the hindbrain, a region that coordinates essential functions such as movement, breathing, and sensory processing. During embryonic development, these progenitors proliferate within organized segments called rhombomeres and respond to patterning signals that regulate regional identity, cell division, migration, and differentiation. Studying hindbrain progenitors helps explain how brainstem and cerebellar circuits form and how disruptions in developmental signaling can contribute to neurological disorders. These cells also provide valuable systems for investigating neural development, modeling disease, and evaluating strategies for tissue repair.

Embryonic neurons are born in the ventricular zone of the neural tube, but migrate to reach appropriate targets. Facial branchiomotor (FBM) neurons are a useful model to study neuronal migration. This protocol describes the wholemount ex vivo culture of mouse embryo hindbrains to investigate mechanisms that regulate FBM migration.

An Antibody Uptake Assay for Imaging Notch/DeltaD Signaling in Zebrafish Radial Glia Progenitors

2025

This video demonstrates an antibody uptake assay to image Notch/DeltaD signaling in zebrafish radial glia progenitors. It involves mounting agarose-embedded dechorionated zebrafish embryos, injecting a dye solution targeting DeltaD ligands on RGPs, and imaging under a fluorescence microscope for asymmetric cell division confirming Notch/DeltaD signaling activation.
