Hippocampal Layers

Hippocampal layers are the organized cellular and fiber zones that form the hippocampus, a brain structure central to memory formation, spatial navigation, and learning. Their arrangement supports directed information flow: entorhinal cortex inputs reach dentate gyrus granule cells through the perforant path, which relay signals to CA3 pyramidal neurons through mossy fibers, while CA3 sends Schaffer collateral projections to CA1. Distinct layers, including the molecular, granule cell, pyramidal cell, and radiatum regions, therefore create a structured hippocampal circuit. Studying this organization helps researchers investigate synaptic plasticity, memory encoding, development, and neurological disorders such as epilepsy and Alzheimer’s disease.

Organotypic Hippocampal Slice Cultures

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.

This article describes the protocol for culturing low-density primary hippocampal neurons growing on glass coverslips inverted over a glial monolayer. The creation of functional microtissues within microfluidic devices requires the stabilization of cell phenotypes by adapting traditional cell culture techniques to the limited spatial dimensions in microdevices. Modification of collagen allows the layer-by-layer deposition of ultrathin collagen assemblies that can stabilize primary cells, such as hepatocytes, as microfluidic tissue models.
