Ledge Test

The Ledge Test is a behavioral neuroscience assay that evaluates balance, motor coordination, and sensorimotor function in rodents by observing how they navigate a narrow, elevated surface. During the test, an animal is placed on a ledge and monitored for its ability to maintain posture, walk along the edge, and recover from slips or instability, with performance commonly assessed through latency, distance, or error counts. Researchers use these measures to identify motor impairments after brain injury, neurodegenerative disease, genetic manipulation, or drug treatment, making the assay useful for characterizing neurological phenotypes and evaluating therapeutic effects.

Rats' anxiety-like behavior tested by the elevated plus maze varies highly depending on the testing environment. Here we show how apparatus settings, especially arm structure, affect behavior on the maze, and we explain a reliable protocol to detect effects of anxiolytic drugs. This report will contribute to anxiety research.

A Simple Composite Phenotype Scoring System for Evaluating Mouse Models of Cerebellar Ataxia

We describe a protocol for the rapid and sensitive quantification of disease severity in mouse models of cerebellar ataxia. Measures include hind limb clasping, ledge test, gait and kyphosis. This protocol effectively discriminates between affected and non-affected individuals, and detects the progression of affected individuals over time.

A Simple Alternative to Stereotactic Injection for Brain Specific Knockdown of miRNA

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.

Here, we present a protocol to use a curvelet transform-based, open-source MATLAB software tool for quantifying fibrillar collagen organization in the extracellular matrix of both normal and diseased tissues. This tool can be applied to images with collagen fibers or other types of line-like structures.
