Microglia Biosensor

A microglia biosensor is a cell-based sensing system that uses microglia, the brain’s resident immune cells, to report biochemical or physical signals associated with neural injury and inflammation. When exposed to stimuli such as pathogen-associated molecules, damaged-cell signals, or inflammatory mediators, microglia alter their morphology, gene expression, cytokine secretion, and other measurable outputs; these responses can be coupled to optical, electrical, or molecular readouts. In neuroscience, microglia biosensors help detect neuroinflammatory activity, evaluate drug effects, and study communication between immune and neural cells, supporting earlier identification of disease-related changes and more physiologically relevant models of brain dysfunction.

Microglia (immune cells of the brain), are used as a surrogate biosensor to determine how nanoparticles influence neurotoxicity. We describe a series of experiments designed to assay microglial response to nanoparticles and exposure of hypothalamic neurons to supernatant from activated microglia to determine neurotoxicity.

Lytic phage biosensors and antibody beads are able to discriminate between methicillin resistant (MRSA) and sensitive staphylococcus bacteria. The phages were immobilized by a Langmuir-Blodgett method onto a surface of a quartz crystal microbalance sensor and worked as broad range staphylococcus probes. Antibody beads recognize MRSA.

To detect healthy cells in whole animals that contain low levels of caspase activity, the highly sensitive biosensor designated CaspaseTracker was generated for Drosophila. Caspase-dependent biosensor activity is detected in long-lived healthy cells throughout the internal organs of adult animals reared under optimized conditions in the absence of death stimuli.
