Mouse Neuron Culture

Mouse neuron culture is a laboratory method for maintaining neurons isolated from mouse nervous tissue under controlled conditions, enabling direct study of neuronal structure and function. Researchers typically dissociate brain or spinal cord tissue, plate the cells on an adhesive substrate, and provide a nutrient-rich medium that supports survival, neurite extension, synapse formation, and electrical activity while limiting contamination from other cell types. In neuroscience, these cultures help investigate neurodevelopment, cellular signaling, neurotoxicity, and disease mechanisms. They also provide a tractable platform for testing drugs, genetic manipulations, and imaging approaches before studies in more complex animal or tissue models.

Primary Culture of Mouse Dopaminergic Neurons

Dopaminergic neurons play a vital regulatory role in the brain. Their loss is associated with Parkinson's disease. In this video, we show how to generate primary cultures of central dopaminergic neurons from embryonic mouse mesencephalon. Such cultures are useful to study the extreme vulnerability of these neurons to various stresses.

A Technique to Culture Mouse Hippocampal Neurons

The video demonstrates a method for culturing neurons from murine hippocampi. Initially, the hippocampi undergo enzymatic digestion to break down the extracellular tissue matrix. Subsequently, the digested tissue is mechanically disrupted to isolate individual neurons. Finally, these neurons are cultured on glial feeder cells.

This video shows a procedure for generating neuronal cultures from late embryo and early postnatal mouse cortex.
