Ribbon Synapses

Ribbon synapses are specialized presynaptic junctions that support continuous, rapid neurotransmitter release in sensory neurons, making them essential for faithfully encoding changing visual, auditory, and vestibular signals. At these synapses, an electron-dense protein structure called the ribbon tethers synaptic vesicles near voltage-gated calcium channels; membrane depolarization opens the channels, and calcium influx triggers vesicle fusion while nearby vesicles are rapidly replenished. Found prominently in retinal photoreceptors, bipolar cells, and inner-ear hair cells, ribbon synapses sustain transmission during prolonged stimulation while preserving temporal precision. Their organization provides a framework for studying sensory coding, synaptic transmission, and disorders affecting vision or hearing.

A protocol for processing young adult and aged gerbil cochleae by immunolabeling the afferent synaptic structures and hair cells, quenching autofluorescence in aged tissue, dissecting and estimating the length of the cochleae, and quantifying the synapses in image stacks obtained with confocal imaging is presented.

This manuscript describes an experimental protocol for evaluating the morphological characteristics and functional status of ribbon synapses in normal mice. The present model is also suitable for noise-induced and age-related cochlear synaptopathy-restricted models. The correlative results of previous mouse studies are also discussed.
