Epicardial Vasospasm

Epicardial vasospasm is a transient, intense constriction of a major coronary artery that reduces myocardial blood flow and can cause ischemic chest pain, even without fixed obstructive disease. It develops when coronary vascular smooth muscle contracts excessively, often in association with endothelial dysfunction and altered vasomotor signaling, producing temporary arterial narrowing and possible ST-segment elevation. In pharmacology, the condition provides a model for studying coronary tone and evaluating vasodilator therapy, including nitrates and calcium channel blockers. Understanding its mechanisms supports diagnosis, prevention of recurrent episodes, and safer assessment of drugs that influence vascular smooth muscle or endothelial function.

Epicardial Outgrowth Culture Assay and Ex Vivo Assessment of Epicardial-derived Cell Migration

Here, we describe methods for isolating primary mouse epicardial cells by an outgrowth culture assay and assessing the functional migration of epicardial-derived cells (EPDC) using an ex vivo heart culture system. These protocols are suitable for identifying genetic and chemical modulators of epicardial epithelial-to-mesenchymal transition (EMT) and motility.

The goal of this article is to present a method that allows a 3-dimensional reconstruction of the cerebrovascular tree in mice after micro computed tomography and determination of volumes of entire vessel segments that can be used to quantify cerebral vasospasm in murine models of subarachnoid hemorrhage.
