Maxam-Gilbert Sequencing

No método Maxam-Gilbert, ou clivagem química, o DNA molde é primeiro desnaturado e radiomarcado na extremidade 5 'com um fósforo 32. 
A reação de sequenciamento é executada em quatro tubos de reação diferentes com quatro produtos químicos diferentes com a capacidade de modificar diferentes nucleotídeos no DNA. 
Por exemplo, o ácido fórmico atacará apenas as purinas, adenina ou guanina, enquanto a hidrazina atacará apenas as pirimidinas - citosina ou timina. Da mesma forma, produtos químicos como o dimetil sulfato atacam apenas um único nucleotídeo.
A piperidina é então usada para clivar o DNA nas bases modificadas, gerando fragmentos de DNA radiomarcados de diferentes comprimentos.

O método Maxam-Gilbert depende do uso de diferentes produtos químicos que podem clivar a sequência de DNA em locais específicos, da separação dos fragmentos de DNA resultantes de tamanho variável usando eletroforese e da decifração da sequência de DNA das bandas de gel resultantes. Desafios do Método Maxam-Gilbert O método Maxam-Gilbert só pode ser usado para resolver normalmente até 400 pb em uma única execução, o que é comparativamente menor do que outros métodos de sequenciamento tradicionais, como o sequenciamento Sanger. Chemical cleavage DNA sequencing uses specific chemicals to cut DNA at known sites. Allan Maxam and Walter Gilbert described this method in the same year that the Sanger sequencing method was discovered. It reads the DNA sequence by breaking purified DNA into fragments and then studying the fragment pattern.

The method depends on chemicals that cleave DNA at specific bases. The resulting fragments are separated by electrophoresis, which is a way to sort DNA pieces by size using an electric field. After that, the sequence is determined from the band pattern seen on the gel.

The Maxam-Gilbert method has important limits. It usually resolves up to about 400 base pairs in one run, which is less than some traditional sequencing methods such as Sanger sequencing. It also needs relatively large amounts of template DNA, and it uses radioisotopes and hazardous chemicals, so it is not ideal for routine use.

Even with these drawbacks, the method has one advantage. Because it works directly with purified DNA, it can be more accurate than Sanger sequencing. Today, it is used less often, but it still has value in some DNA fingerprinting methods and in DNA structural studies.

EXPLANATION: The goal is to help students appreciate that the basic science knowledge being studied can be manipulated and used to design useful tools for further scientific investigations, medical treatments, agricultural yields, etc. A detailed understanding of any technical procedures is not expected, but rather a "big picture" view of concepts the technology utilizes and the applications the technology is being used for. These technologies may be best investigated along with the standards from LS1 From Molecules to Organisms: Structure and Functions, as the students are learning about DNA and protein. Comparative DNA and protein sequence analysis is also a large part of current phylogenic research and could accompany LS4 Biological Change: Unity and Diversity. Students may also consider the pros and cons of biotechnical applications. Investigation of techniques utilized could come from field trips to witness the use of these techniques in practice, lab exercises, virtual labs, simulation activities, interviews with professionals, etc. Molecular techniques could include: PCR, electrophoresis, restriction enzyme digestion of DNA, DNA sequencing, plasmid-based transformation, transfection, etc. These techniques are used in fields of medicine, agriculture, biomedical engineering, forensic science, etc.
