Cyclin Concentration Fluctuation

Cyclin concentration fluctuation is the periodic rise and fall of cyclin protein levels that helps coordinate progression through the cell cycle. These changes occur when cells regulate cyclin gene expression, translation, and targeted protein degradation; accumulating cyclins activate cyclin-dependent kinases (CDKs), while ubiquitin-mediated destruction lowers CDK activity and permits transitions to the next phase. In biology, measuring these oscillations helps researchers connect molecular timing with DNA replication, mitosis, and checkpoint control. Studying cyclin dynamics also supports investigations of abnormal cell proliferation, including how disrupted regulation can contribute to cancer and how timing-based interventions might affect dividing cells.

Seasonal Fluctuations

2026

Seasonal fluctuations refer to regular and predictable changes in economic activity that happen at specific times each year. These variations arise from recurring influences such as weather conditions, holidays, and institutional schedules. Unlike business cycles, which are irregular, seasonal fluctuations are predictable and happen every year.Weather patterns can affect the economy. Certain industries, such as agriculture and construction, tend to experience higher or lower activity during...

Fluorescence Fluctuation Spectroscopy to Study Protein Homo-Oligomerization

2025

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.

Fluorescence Fluctuation Spectroscopy to Study Protein Interaction at Cell Contacts

2025

This video demonstrates the use of fluorescence fluctuation spectroscopy to detect the interaction among cell surface proteins at cell-cell contacts. By expressing the transmembrane adhesion receptor of interest labeled with a fluorescent protein and mixing two different cell populations harboring two spectrally separated fluorescent labels, the trans-interaction between the receptors of two neighboring cells with different-colored fluorescence is assessed via cross-correlation in the...

Positive Regulator Molecules

2019

To consistently produce healthy cells, the cell cycle—the process that generates daughter cells—must be precisely regulated. Internal regulatory checkpoints ensure that a cell’s size, energy reserves, and DNA quality and completeness are sufficient to advance through the cell cycle. At these checkpoints, positive and negative regulators promote or inhibit a cell’s continuation through the cell cycle. Positive regulators include two protein groups that allow cells to pass through regulatory...
