Brain Metastases Model

A brain metastases model is an experimental system that reproduces how cancer cells spread to, survive in, and grow within the brain, enabling study of this clinically challenging form of cancer progression. These models may use cultured cells, brain organoids, animal systems, or patient-derived tumors to examine processes such as blood-brain barrier crossing, interactions with neural and stromal cells, and metastatic colonization. Researchers use them to investigate tumor biology, identify molecular drivers, and evaluate how treatments reach and affect metastatic lesions. Improved models can better reflect the brain microenvironment, strengthen preclinical drug testing, and support the development of more effective therapies for patients with advanced cancer.

Intracranial brain metastasis modeling is complicated by an inability to monitor tumor size and response to treatment with precise and timely methods. The presented methodology couples intracranial tumor injection with magnetic resonance imaging analysis, which when combined, cultivates precise and consistent injections, enhanced animal monitoring, and accurate tumor volume measurements.

This video describes the protocol for generating metastases by the PDX derived CRC organoids labeled with GFP lentivirus into the spleen of an immunodeficient mouse. Cancer micrometastases colonizing other organs can be detected using a fluorescence microscope to assess the GFP expression.

A method is presented to measure microcirculatory blood flow velocity in pulmonary cancer metastases of the pleural surface in rats in an automated fashion, using closed-chest pulmonary intravital microscopy. This model has potential to be used as a widespread tool to perform physiologic research on pulmonary metastases in rodents.
