Human Colonic Organoids

Human colonic organoids are three-dimensional, stem cell-derived cultures that reproduce key structural and functional features of the human colon, making them valuable models for studying intestinal development. In a defined extracellular matrix, intestinal stem cells self-renew and differentiate into epithelial lineages as controlled signals, including Wnt, R-spondin, EGF, and BMP pathway modulation, regulate tissue organization and cell fate. In developmental biology, these organoids provide experimentally accessible systems for examining crypt formation, epithelial maturation, and congenital or disease-associated defects, while also supporting studies of host-microbe interactions, drug responses, and patient-specific tissue behavior.

An In Vitro Method to Generate Human Brain Organoids

The video demonstrates a method for generating human brain organoids from induced pluripotent stem cells. It shows how the neurosphere forms a neuroectodermal layer, which suppresses the formation of other germ layers. The neurospheres are embedded in a 3D gel matrix and form neuroepithelial buds. When incubated with organoid media, these neuroepithelial buds differentiate into specific brain regions, resulting in the formation of mature brain organoids.
