Safranin Counterstain

Safranin counterstain is a secondary dye used in differential staining to provide contrast after a primary stain and, when needed, a decolorization step. In the Gram staining process, alcohol or acetone removes the crystal violet–iodine complex from bacteria with thinner peptidoglycan layers, allowing safranin to color them pink or red, while Gram-positive cells remain purple. This contrast helps researchers distinguish bacterial groups in environmental samples such as soil, water, and biofilms, supporting microbial identification and assessment of community composition. Because results depend on smear quality, reagent timing, and decolorization, consistent technique is essential for reliable microscopy.
