Hemolymph Analysis

Hemolymph analysis is the examination of the circulating fluid in arthropods and other invertebrates to assess cellular and biochemical features associated with physiology, immunity, and infection. The analysis typically involves collecting hemolymph, characterizing hemocytes by morphology or cell counts, and measuring soluble immune factors such as antimicrobial activity, phenoloxidase responses, or pathogen-associated changes. In immunology and infection research, these measurements reveal how innate defenses detect and respond to microbes, distinguish cellular from humoral responses, and reflect disease progression or host condition. Hemolymph analysis supports studies of host-pathogen interactions, vector competence, insect immunity, and the effects of environmental or experimental treatments.

In this protocol we demonstrate how to collect hemolymph and salivary glands from feeding Ixodes scapularis nymphs. We also demonstrate saliva collection from female I. scapularis adults.
