Human T Cell Quantification

Human T cell quantification is the measurement of T lymphocyte abundance in blood or other biological samples, providing a basis for evaluating immune status and responses to infection. Commonly, fluorescent antibodies label markers such as CD3, CD4, and CD8, while flow cytometry identifies and counts cells through sequential gating; counting beads or sample-volume measurements can convert frequencies into absolute cell numbers. These measurements help characterize immune dysfunction, compare cellular responses during infection, monitor treatment or vaccination, and distinguish changes in helper and cytotoxic T cell populations. Reliable quantification supports immunological research and clinical assessment of disease progression and recovery.

This video demonstrates the quantification of inflammatory mediators during infection by incubating human lung-tissue-derived immune cells with Haemophilus influenzae. Flow cytometry is used to measure cytokine fluorescence in labeled lymphocytes, correlating with the production of pro-inflammatory cytokines in response to bacterial infection.

We describe a method to evaluate the effect of pre-existing immunity against dengue virus on the Zika virus infection by using human serum, primary human cells, and infection quantification by quantitative real-time polymerase chain reaction.
