Egfp Bacteria Coincubation

EGFP bacteria coincubation is a laboratory method that places bacteria engineered to express enhanced green fluorescent protein (EGFP) together with immune or host cells to visualize and study their interactions. EGFP produces green fluorescence when excited with the appropriate light, allowing researchers to track bacterial attachment, uptake, localization, and persistence during controlled coculture conditions. In immunology and infection research, this approach helps assess how cells recognize and respond to bacteria, including changes in phagocytosis and intracellular handling. Fluorescent labeling supports microscopy, flow cytometry, and quantitative comparisons of host-pathogen interactions, providing a direct way to investigate cellular infection mechanisms.

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.

By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
