Coronavirus Spike Protein

The coronavirus spike protein is a trimeric glycoprotein on the viral surface that mediates host-cell entry and serves as a major target of antiviral immunity. Its receptor-binding regions attach to specific molecules on susceptible cells, while proteolytic cleavage and structural rearrangements expose fusion machinery that merges viral and cellular membranes. In immunology and infection research, spike protein is used to study viral tropism, neutralizing antibodies, vaccine responses, and immune escape. Comparing spike sequences and structures across variants also helps researchers assess changes in receptor binding, transmissibility, and the effectiveness of vaccines or antibody-based interventions.

We present a fluorogenic peptide cleavage assay that allows a rapid screening of the proteolytic activity of proteases on peptides representing the cleavage site of viral fusion peptides. This method can also be used on any other amino acid motif within a protein sequence to test for the protease activity.

In this video, we demonstrate an alternative method for detection and titering of viruses using an enzymatic antigen detection technique known as an immunoperoxidase assay. Here, we will show you how to collect your viral samples, prepare the cells for testing, and finally the immunoperoxidase assay using serial dilutions to determine the viral titer.

Coronaviruses, including the severe acute respiratory syndrome coronavirus (SARS-CoV), are enveloped viruses characterized by their single-stranded, positive-sense RNA genome and helical nucleocapsid structure. The hallmark of these viruses is their club-shaped spike (S) glycoproteins that protrude from the viral envelope, facilitating attachment to host cells. Typically, coronaviruses infect the upper respiratory tract, often causing mild or asymptomatic disease. However, certain strains like...
