Athymic Nude Rats

Athymic nude rats are immunodeficient laboratory rodents that lack a functional thymus and therefore provide a controlled model for studying human disease and tissue responses. Their inherited nude phenotype prevents normal thymic development and T-lymphocyte maturation, reducing cell-mediated immunity while permitting many transplanted human cells or tissues to survive without immediate rejection. In medicine, researchers use these rats for human tumor xenografts, transplantation studies, drug evaluation, and regenerative research, where tumor growth, treatment response, graft integration, and disease mechanisms can be measured in vivo; their increased susceptibility to infection also requires careful husbandry and experimental controls.

Optimized Protocols for Mycobacterium leprae Strain Management: Frozen Stock Preservation and Maintenance in Athymic Nude Mice

Mycobacterium leprae, the causative agent of leprosy, does not grow in vitro. We describe an easy to follow protocol to prepare a bacillary suspension to ensure the maintenance of large quantities of M. leprae for a variety of applications. Protocols for propagation by mouse footpad inoculation, evaluation of viability, freezing and thawing bacillary stock are described in detail.

Animal models are important tools for the evaluation of tissue-engineered grafts. This paper presents the protocol for preparing an electrospun biodegradable polymer graft for use in anterior cruciate ligament tissue engineering, as well as a surgical protocol for implantation in a rat model.

In the pathogenesis of bone metastasis, angiogenesis is a crucial process and therefore represents a target for imaging and therapy. Here, we present a rat model of site-specific breast cancer bone metastasis and describe strategies to non-invasively image angiogenesis in vivo using magnetic resonance imaging, volumetric computed tomography and ultrasound.

Fat grafting is an essential technique for reconstructing soft tissue deficits. However, it remains an unpredictable procedure characterized by variable graft survival. Our goal was to devise a mouse model that utilizes a novel imaging method to compare volume retention between differing techniques of fat graft preparation and delivery.
