Disturbed Flow Model

The Disturbed Flow Model is an experimental framework that simulates irregular, oscillatory, or low shear stress patterns in blood vessels to study how abnormal flow affects vascular biology. By exposing endothelial cells or vessel tissues to defined flow conditions, researchers reproduce the mechanical forces found near arterial branches and curves, where altered signaling can increase endothelial permeability, inflammatory gene expression, and leukocyte adhesion. In medicine, this model helps clarify how disturbed hemodynamics contribute to atherosclerosis and other vascular disorders. It also supports evaluation of molecular targets, therapeutic strategies, and factors that promote endothelial protection and cardiovascular health.

This describes a partial carotid ligation surgery, which causes disturbed flow conditions and subsequent atherosclerosis development (in two weeks) with intraplaque neo-vascularization (in four weeks) in the mouse common carotid artery. We also describe a novel method of RNA isolation from the carotid intima, providing high purity endothelial RNA.

Three-dimensional flow separation induced by a wall-mounted model polyp and its impact on the wall pressure loading are examined using particle image velocimetry, skin friction line visualization, and wall pressure measurements.
