Preclinical Lung Model

A preclinical lung model is an experimental system that reproduces key features of lung structure, function, or disease before a therapy or intervention reaches clinical testing. These models may use cultured airway or alveolar cells, engineered tissue, organoids, or animal systems to examine processes such as epithelial injury, inflammation, gas-exchange behavior, and responses to candidate treatments under controlled conditions. In medicine, preclinical lung models help researchers investigate respiratory disease mechanisms, evaluate drug efficacy and toxicity, and refine therapeutic strategies while reducing uncertainty before human studies. Their predictive value depends on how closely the model reflects human lung biology and the clinical condition being studied.

Implementation of an orthotopic model of renal cell carcinoma in immunocompetent mice affords the investigator a clinically-relevant system defined by the presence of a primary renal tumor and lung metastases in the same animal. This system can be used to preclinically test a variety of treatments in vivo.

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.
