FISH - Fluorescent In-situ Hybridization

Флуоресцентная гибридизация in situ или FISH — это метод, используемый для обнаружения расположения определенных генов или частей генов на хромосомах в фиксированных клетках.
В этом методе флуоресцентно меченая одноцепочечная ДНК используется в качестве зонда для связывания частей хромосом, содержащих комплементарную последовательность ДНК, называемых мишенью.
Во-первых, клетки задерживаются в интерфазе или метафазе. Арестованные клетки суспензируют в буферном растворе и выкладывают на чистое предметное стекло.
Затем целевая ДНК и помеченные зонды денатурируются с помощью тепла или химических веществ. На этом этапе двухцепочечная ДНК отделяется, оставляя зонд и целевую ДНК в виде одноцепочечных последовательностей, способных к спариванию оснований.
Затем зонд и целевую ДНК смешивают вместе и инкубируют в течение ночи. Это позволяет зонду гибридизоваться с его комплементарной последовательностью на хромосомах.
После завершения гибридизации излишки зонда смываются.
Если зонд был помечен флуорофором, места расположения гибридизированной ДНК можно непосредственно визуализировать с помощью флуоресцентного микроскопа.
В противном случае гибридизованная ДНК должна быть флуоресцентно помечена перед визуализацией. Для этого последовательность зонда должна содержать модифицированный нуклеотид, связанный с гаптеном – небольшой молекулой, к которой флуорофор, например, флуоресцентное антитело, может прикрепляться.
Такая прямая визуализация генов очень полезна для клеточных диагностических анализов, где генетические аномалии могут быть обнаружены в результате любой потери..., приобретения..., или перегруппировки хромосом во время деления клетки.

Флуоресцентная гибридизация in situ, или FISH, была разработана в начале 1980-х годов и быстро стала одним из наиболее широко используемых методов цитогенетики. Меченые зонды используются для связывания комплементарных последовательностей ДНК или РНК на хромосоме или в области внутри клетки. Раньше зонды можно было получить только путем клонирования или обратной транскрипции ДНК-матрицы. В настоящее время олигонуклеотиды-зонды можно синтезировать синтетически. Кроме того, благодаря развитию оптических методов, ПЗС-камер и сложного программного обеспечения для обработки изображений сигналы FISH можно интерпретировать с большей чувствительностью и высокой специфичностью. FISH is a DNA and RNA detection method used in cytogenetics and medical testing. FISH stands for fluorescence in situ hybridization. It uses labeled probes that bind to complementary DNA or RNA sequences on a chromosome or inside a cell.

The method was developed in the early 1980s and became widely used because it can show specific genetic targets with high detail. In the past, probes were made by cloning or by reverse transcription of a DNA template. Today, probe oligonucleotides can also be synthesized synthetically.

Better optical tools have improved FISH results. CCD cameras and image-processing software help scientists read FISH signals with greater sensitivity and high specificity. These advances expanded FISH from gene mapping and cytogenetics into the diagnosis of genetic and infectious diseases.

FISH images can reveal chromosomal aberrations, or chromosome changes. These include common aneuploidy, which means an abnormal number of chromosomes in a cell, as well as microdeletion and microduplication syndromes, where a small chromosome region is lost or gained. FISH can also help identify subtelomeric rearrangements.

FISH is also used to detect microbial pathogens in human blood cells. In these tests, short fluorescence-labeled DNA binds to microbial 16s rRNA and can be seen with a fluorescence microscope. This helps identify infectious agents at the genus or species level.

Because FISH gives rapid results, it is useful for finding slow-growing organisms such as Mycobacteria, the bacteria that cause tuberculosis. It also helps detect microbes that are difficult to culture, such as Tropheryma whipplei. For this reason, FISH-based diagnostics are important in both genetics and infection testing.

Investigation of techniques utilized could come from field trips to witness the use of these techniques in practice, lab exercises, virtual labs, simulation activities, interviews with professionals, etc. Molecular techniques could include: PCR, electrophoresis, restriction enzyme digestion of DNA, DNA sequencing, plasmid-based transformation, transfection, etc. These techniques are used in fields of medicine, agriculture, biomedical engineering, forensic science, etc.

EXPLANATION: The utilization of new technologies in any field of science is dependent on both economic and social factors. In addition to evaluating these factors, scientists must also consider long-term consequences that may not be initially apparent. The emphasis should be on the construction of a rationale argument that supports a position on the use of an application with ethical and social impact. Students should begin to appreciate the differences in ethical values that exist, and recognize that discussion of these values is imperative as knowledge and technology continue to advance, even when resolutions of differences can be rare. After investigation of a specific biotechnology application, students can write a position paper and/or participate in a classroom debate.
