Криоэлектронная микроскопия или крио-ЭМ исследует замороженные биологические образцы при криогенных температурах ниже −150 °С.

Высокоэнергетические электроны, используемые в ЭМ, могут повредить химические связи образца. Замораживание образца помогает предотвратить это повреждение, позволяя получать изображения с разрешением, близким к атомному.
В крио-ЭМ небольшие биологические образцы в растворе остекловываются, то есть быстро замораживаются в жидком этане. Витрификация фиксирует образец в его исходном состоянии в тонком слое воды, и вода в образце образует стекловидное состояние без появления кристаллов льда.
Более толстые образцы, такие как клетки и ткани, не могут быть витрифицированы в тонком жидком слое. Таким образом, эти образцы могут быть исследованы с помощью метода крио-ЭМ срезов стекловидного тела. В этом методе образец сначала остекловывается путем замораживания под высоким давлением, а затем разрезается на ультратонкие срезы при температуре −140 °C перед наблюдением с помощью крио-ЭМ.

Cryo-electron Microscopy

С другой стороны, криоЭМ позволяет изучать более крупные белки, мембраносвязанные рецепторы или биомолекулярные комплексы. 

 Крио-ЭМ широко используется в биохимии для изучения структуры, функций и взаимодействия биомолекул. Инфекционные агенты, такие как вирусы, можно идентифицировать и изучать с помощью криоЭМ. Например, во время вспышки вируса Зика в Бразилии крио-ЭМ использовалась для создания трехмерного изображения структуры вируса, чтобы можно было идентифицировать потенциальные мишени противовирусных препаратов.

 <h4>Cryo-electron Microscopy</h4>

 Conventional electron microscopy (EM) involves dehydration, fixation, and staining of biological samples, which distorts the native state of biological molecules and results in several artifacts. Also, the high-energy electron beam damages the sample and makes it difficult to obtain high-resolution images. These issues can be addressed using cryo-EM, which uses frozen samples and gentler electron beams. The technique was developed by Jacques Dubochet, Joachim Frank, and Richard Henderson, for which they won the Nobel Prize in Chemistry in 2017.

 X-ray diffraction and nuclear magnetic resonance (NMR) techniques are also used for obtaining high-resolution structures of biomolecules or studying their conformational changes. However, to get the X-ray structure, a molecule needs to be crystallized, which may not be possible every time. Even if a molecule is crystallizable, the crystallization process can alter its biomolecular structure, which is no longer representative of the native structure. Cryo-EM does not require a crystallized sample, and also enables visualization of molecular movements or interactions of biomolecules in their native state. Similarly, NMR technique is only limited to relatively small soluble proteins and is not suited for the non-polar proteins embedded in cell membranes. On the other hand, cryo-EM enables the study of larger proteins, membrane-bound receptors, or biomolecular complexes.

 Cryo-EM is extensively being used in biochemistry to study the structure, function, and interaction of biomolecules. The infectious agents, such as viruses, can be identified and studied using cryo-EM. For example, during the Zika virus outbreak in Brazil, cryo-EM was utilized to create a 3D image of the virus structure so that the potential anti-viral drug targets could be identified.

Cryo-EM, or cryo-electron microscopy, lets scientists image biomolecules in their frozen, native state. It avoids the dehydration, fixation, and staining used in conventional electron microscopy, which can distort samples and create artifacts. It also reduces damage from the electron beam by using gentler imaging conditions.

This method is useful when other structure tools have limits. X-ray diffraction needs a molecule to form a crystal, and that is not always possible. Even when crystallization works, it can change the biomolecule’s structure so it no longer matches the native form.

Cryo-EM also fills gaps left by nuclear magnetic resonance, or NMR. NMR is mainly used for relatively small soluble proteins and is not suited to non-polar proteins in cell membranes. Cryo-EM can be used to study larger proteins, membrane-bound receptors, and biomolecular complexes.

Researchers use cryo-EM to study biomolecule structure, function, and interactions. It can also help identify and analyze infectious agents such as viruses. During the Zika virus outbreak in Brazil, cryo-EM was used to build a 3D image of the virus so potential antiviral drug targets could be found.
