Dms Probing

DMS probing is a chemical method for mapping RNA structure by identifying nucleotide positions that remain accessible to dimethyl sulfate, providing insight into RNA folding and interactions. Dimethyl sulfate preferentially methylates exposed adenine and cytosine bases, while nucleotides constrained by base pairing or protein contacts are less reactive; reverse transcription then detects modification sites through stops or mutations in the resulting cDNA. In biochemistry, this approach helps characterize secondary structure, RNA conformational changes, ligand binding, and ribonucleoprotein assembly. Its compatibility with cellular samples also supports analysis of RNA structure in native biological environments.
