P53 Aggregation

p53 aggregation is the abnormal self-association of misfolded p53 tumor-suppressor proteins into oligomers and amyloid-like fibrils, a process that can compromise a central defense against cancer. Normally, p53 forms functional tetramers that bind DNA and activate genes controlling cell-cycle arrest, DNA repair, senescence, and apoptosis; mutations or cellular conditions that destabilize its structure can promote aggregation and reduce these activities. Aggregated p53 may also sequester functional p53 or related proteins, further weakening tumor-suppressive responses. Studying this process helps clarify how specific p53 mutations contribute to tumor development and supports research into aggregation inhibitors, diagnostic biomarkers, and strategies for restoring p53 function.

Hs578T breast cancer cells harboring the p53 V157F mutation exhibit significantly higher Thioflavin T fluorescence compared to MCF7 cells, indicating enhanced protein aggregation. Multipoint fluorescence measurements improve detection accuracy and reliability in identifying β-sheet-rich aggregates, underscoring the importance of aggregation-prone p53 mutations in cancer research and the development of therapeutic strategies.

The goal of the method presented here is to explore protein aggregation during normal aging in the model organism C. elegans. The protocol represents a powerful tool to study the highly insoluble large aggregates that form with age and to determine how changes in proteostasis impact protein aggregation.
