Rat Spine Dissection

Rat spine dissection is an anatomical technique used to expose and examine the vertebral column, spinal cord, and surrounding tissues in a laboratory animal, providing a structured view of tissues relevant to disease research. The procedure typically involves carefully removing overlying muscles and connective tissues, isolating spinal structures, and preserving anatomical relationships for observation or tissue collection. In cancer research, this approach supports investigation of tumor growth near the spine, metastatic involvement of bone or neural tissues, and treatment-related changes. It can also provide tissue samples for histology, molecular analysis, and evaluation of disease progression in preclinical models.

This article describes a working protocol to image dendritic spines from hippocampal neurons in vitro using Structured Illumination Microscopy (SIM). Super-resolution microscopy using SIM provides image resolution significantly beyond the light diffraction limit in all three spatial dimensions, allowing the imaging of individual dendritic spines with improved detail.

Source: Gouder, et. al. Three-dimensional Quantification of Dendritic Spines from Pyramidal Neurons Derived from Human Induced Pluripotent Stem Cells. J. Vis. Exp. (2015).This video demonstrates the imaging and analysis of dendritic spines in transduced, immunolabeled pyramidal neurons using confocal microscopy. Background noise is reduced, and dendrites are traced by estimating their diameter and length. Automated spine segmentation involves setting parameters for dendrite and spine dimensions...
