Capillary Electrode Preparation

Capillary electrode preparation is the fabrication of small electrochemical electrodes within narrow glass or polymer capillaries, enabling controlled measurements in limited sample volumes. A conductive wire or filament is positioned inside the capillary, secured and electrically insulated, then exposed at the tip and polished to create a defined working surface; the capillary walls confine the conductor and help minimize unwanted electrical contact. Proper preparation produces a stable, reproducible electrode geometry for techniques such as voltammetry and amperometry. These electrodes support localized chemical analysis, microvolume experiments, and studies requiring precise control of the electrode–solution interface.

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.
