Retinal Axons

Retinal axons are the long projections of retinal ganglion cells that transmit visual information from the eye to the brain, making them a key model for studying neural circuit formation. During development, their growth cones extend through the optic nerve and respond to molecular guidance cues that direct axons toward appropriate targets, determine whether they cross at the optic chiasm, and establish organized maps in visual regions such as the superior colliculus. Subsequent activity-dependent refinement strengthens useful connections and eliminates others. Research on retinal axons clarifies mechanisms of visual system development and informs studies of congenital disorders, axon regeneration, and neural repair.

We demonstrate an in vivo electroporation protocol for transfecting single or small clusters of retinal ganglion cells (RGCs) and other retinal cell types in postnatal mice over a wide range of ages. The ability to label and genetically manipulate postnatal RGCs in vivo is a powerful tool for developmental studies.
