Vascular Perfusion

Vascular perfusion is the movement of blood or a blood-like fluid through a tissue’s vascular network, delivering oxygen and nutrients while removing metabolic waste. It depends on pressure gradients generated by the heart or an external pump, along with vessel diameter, resistance, branching patterns, and endothelial permeability that regulate fluid flow and exchange. In developmental biology, studying vascular perfusion reveals how blood vessels form, mature, and integrate with growing organs, while perfusion-based imaging and culture systems help assess tissue function, embryonic development, and engineered vascular networks. These approaches also clarify how impaired circulation contributes to developmental abnormalities and disease.

The protocol describes how to engineer a perfusable vascular network in a spheroid. The spheroid's surrounding microenvironment is devised to induce angiogenesis and connect the spheroid to the microchannels in a microfluidic device. The method allows the perfusion of the spheroid, which is a long-awaited technique in three-dimensional cultures.

Presented is a method for air-inflation with vascular perfusion-fixation of the lungs that preserves the location of cells within airways, alveoli and interstitium for structure-function analyses. Constant airway pressure is maintained with an air-inflation chamber while fixative is perfused via the right ventricle. Lungs are processed for histologic studies.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.
