Gamma-h2ax Immunolabeling

Gamma-H2AX immunolabeling is a fluorescence-based method for detecting phosphorylated histone H2AX, a widely used marker of DNA double-strand breaks and genome instability. When DNA damage occurs, kinases such as ATM phosphorylate H2AX at serine 139, producing γ-H2AX around damaged chromatin; antibodies against this modification then reveal nuclear foci by immunofluorescence microscopy. In genetics research, the technique helps quantify DNA damage and repair, assess chromosome stability, and compare cellular responses to radiation, chemicals, or mutations. Because γ-H2AX foci can be measured at the single-cell level, immunolabeling supports studies of genotoxicity, replication stress, and disease-associated defects in genome maintenance.

We describe here an isolation method to obtain human endocervical intraepithelial lymphocytes for the analysis of intraepithelial gamma delta T cells. This protocol can be extended for the purification of endocervical gamma delta T cells by magnetic beads or by cell sorting.
