Mirna Target Validation

miRNA target validation is the experimental process of confirming that a microRNA directly regulates a specific messenger RNA, rather than merely being computationally predicted to interact with it. Researchers typically test whether the miRNA seed region binds a complementary sequence, often in the mRNA 3′ untranslated region, using reporter assays with wild-type and mutated target sites, alongside measurements of transcript or protein levels. In genetics, validated targets help explain post-transcriptional gene regulation, connect miRNAs to developmental or disease-associated pathways, and distinguish direct regulatory effects from secondary changes. These findings strengthen gene-network models and can guide studies of biomarkers and therapeutic targets.

Biotin-based Pulldown Assay to Validate mRNA Targets of Cellular miRNAs

This report describes a fast and reliable method for validating mRNA targets of cellular miRNAs. The method uses synthetic biotinylated Locked Nucleic Acid (LNA)-based miRNA mimics to capture target mRNA. Subsequently, streptavidin-coated magnetic beads are employed to pulldown the target mRNA for quantification by qPCR polymerase chain reaction.
