Ctl Expansion Protocol

A CTL expansion protocol is a laboratory method for increasing the number of cytotoxic T lymphocytes, immune cells that recognize and destroy infected or abnormal target cells. The process typically stimulates T cells through antigen recognition and co-stimulatory signals, then supports their proliferation with growth-promoting cytokines under controlled culture conditions. Expanded CTLs can be evaluated for phenotype, viability, and target-cell killing, making this approach useful for studying antiviral and antitumor immunity, modeling cellular immune responses, and developing adoptive immunotherapies. In immunology and infection research, the protocol helps connect T-cell activation with measurable effector function.

A new DC independent method for induction and expansion of antigen-specific T cells is described. HLA A2-Ig based artificial Antigen Presenting Cells (aAPC) are loaded with HLA-A2 restricted peptides to efficiently expand CTL of diverse antigen specificity. This technology holds great potential for CTL-based adoptive immunotherapy.

Here, we present a detailed protocol combining cryo-fixation and expansion microscopy methods (Cryo-ExM), allowing for high-resolution imaging with structural preservation adapted to various biological samples.
