Neutralizing Capacity Measurement

Neutralizing capacity measurement is the quantitative assessment of how effectively antibodies or other immune factors block a pathogen, toxin, or viral entry into susceptible cells. In immunology and infection research, serially diluted serum or purified antibodies are mixed with a target agent and applied to cells, with reduced infection, cytopathic effect, or reporter signal indicating neutralization; results are commonly summarized as an inhibitory or neutralizing titer. These measurements help evaluate immune protection, compare vaccine- or infection-induced responses, and characterize antibody activity against pathogen variants. They also support therapeutic antibody development and surveillance of changes in population-level immunity.

We designed a cell-free receptor binding assay in order to estimate the binding of granulocyte-macrophage colony-stimulating factor (GM-CSF) to the receptors. It enables us to evaluate competitive inhibition of biotinylated GM-CSF binding to soluble GM-CSF receptor alpha by GM-CSF autoantibody with excellent reproducibility.
