Dendritic Cell Trafficking

Dendritic cell trafficking is the regulated movement of antigen-presenting dendritic cells between tissues, lymphatic vessels, and lymph nodes, where it links innate detection with adaptive immune activation. Tissue signals such as microbial products and inflammatory cytokines induce chemokine receptors and adhesion molecules that guide dendritic cells through the extracellular matrix, across lymphatic endothelium, and toward lymph nodes; there, they present processed antigen to naïve T cells. Studying this process clarifies how immune responses begin and are coordinated during infection. It also informs vaccine design, cancer immunotherapy, and strategies for controlling chronic inflammation and autoimmune disease.

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.

T Cells Capture Bacteria by Transinfection from Dendritic Cells

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.

Here we describe a novel assay for monitoring prion uptake and trafficking by immune cells immediately following intraperitoneal inoculation by purifying and fluorescently labeling aggregated prion rods from infected brain material then monitoring their uptake and movement from the injection site and characterizing the cells mediating these events.
