Flow Cytometry Assay

A flow cytometry assay is a laboratory method that rapidly measures physical and fluorescent characteristics of individual cells as they pass through a laser-illuminated fluid stream, making it valuable for analyzing heterogeneous cell populations. The instrument detects light scattering and fluorescence from labeled antibodies or other probes, allowing researchers to identify cell types, quantify surface or intracellular markers, and assess cell viability or activation. In immunology and infection research, flow cytometry assays characterize immune responses, detect infected or altered cells, and evaluate changes in cellular phenotype. Multiparameter analysis can reveal relationships among immune-cell subsets and support diagnostics, vaccine studies, and therapeutic development.

Automation of the Micronucleus Assay Using Imaging Flow Cytometry and Artificial Intelligence

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

Over the recent years, live cell-based assays have been used successfully to detect antibodies against surface and conformational antigens. Here, we describe a method using high-throughput flow cytometry enabling the analysis of large cohorts of patients. Detection of novel antibodies will improve diagnosis and treatment of immune-mediated disorders.

This protocol describes a sensitive, cell-based cytotoxicity assay. By enumerating the decrease in frequency of live target CD4+ T cells in the presence of an increasing number of effector CD8+ T cells, this assay allows for the direct assessment of cytolytic activity of antigen-specific CD8+ T cells.
