Immunoglobulin Binding

Immunoglobulin binding is the selective interaction of antibodies with antigens, a fundamental process in immune recognition and defense against infection. The antibody’s variable region binds a specific epitope through complementary shape, charge, and noncovalent interactions, while its Fc region can engage Fc receptors or activate complement to recruit immune effector functions. These interactions may neutralize pathogens and toxins, promote opsonization and phagocytosis, or support antigen detection in laboratory assays. Studying immunoglobulin binding helps researchers evaluate immune responses, characterize antibody specificity and affinity, develop diagnostic tests, and design vaccines or antibody-based therapies.

A Paper-Based Immunoassay for the Detection of Immunoglobulin G

2025

The video demonstrates a paper-based immunoassay, involving amine-functionalized cellulose discs cross-linked with glutaraldehyde. Covalent immobilization of IgG-Fc-specific capture antibodies, followed by the introduction of immunoglobulin G and horseradish peroxidase-conjugated detection antibodies, results in a color change confirming the presence of immunoglobulin G.

Measuring Immunoglobulin G Levels Against a Test Vaccine in a Mouse Serum

2025

This video demonstrates a method for assessing the antibody immune response to a novel nanoemulsion adjuvant vaccine in pre-immunized mice. Immunization enhances antibody response as confirmed by determining the IgG antibody titers.

A Microfluidic Cartridge-Based Detection System for Allergen-Specific Immunoglobulin E

2025

This video demonstrates a protocol for detecting serum-specific immunoglobulin E using a microfluidic cartridge-based chemiluminescence system. Serum IgEs interact with distinct allergens immobilized in the reaction zone of the cartridge, which are subsequently detected with peroxidase-conjugated anti-IgE antibodies and a chemiluminescent substrate, allowing simultaneous detection of multiple allergen-specific IgEs.

An Assay to Detect Serum Anti-aquaporin-4 Immunoglobulin G

2025

The video demonstrates a cell-based assay for detecting anti-aquaporin-4 immunoglobulin G using a biochip containing untransfected cells and transfected cells expressing aquaporin-4. This method has applications in diagnosing neuromyelitis optica spectrum disorders, confirmed by strong green fluorescence on the biochip.

The Equilibrium Binding Constant and Binding Strength

2020

The equilibrium binding constant (Kb) quantifies the strength of a protein-ligand interaction. Kb can be calculated as follows when the reaction is at equilibrium: where P and L are the unbound protein and ligand, respectively, and PL is the protein-ligand complex. As the amount of bound ligand is also related to the rate of ligand binding, experiments can also determine Kb by examining the rates of protein-ligand association (kon) and dissociation (koff) using the following ratio: Thus,...
