Mouse Liver Repopulation

Mouse liver repopulation is an experimental process in which transplanted or genetically introduced hepatocytes replace a substantial portion of a recipient mouse’s liver, providing a model of cell engraftment, tissue regeneration, and liver disease. It works by creating a selective growth advantage for donor cells, often through endogenous hepatocyte injury or loss, allowing the introduced cells to engraft, proliferate, and progressively restore liver mass and function. In immunology and infection research, this system helps assess immune-mediated tissue damage, host responses to infected or modified hepatocytes, and the performance of cell-based therapies. It also supports studies of hepatic regeneration and therapeutic strategies for inherited liver disorders.

This protocol describes decellularization of Sprague Dawley rat kidneys by antegrade perfusion of detergents through the vasculature, producing acellular renal extracellular matrices that serve as templates for repopulation with human renal epithelial cells. Recellularization and use of the resazurin perfusion assay to monitor growth is performed within specially-designed perfusion bioreactors.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
