One Dimensional Tlc

One-dimensional thin-layer chromatography (TLC) is a planar separation method that resolves compounds along a single direction on a coated plate, providing a rapid way to examine complex mixtures. A sample is applied near one edge, and a solvent moves through the stationary phase by capillary action; compounds travel different distances according to their interactions with the adsorbent and their solubility in the mobile phase. In immunology and infection research, one-dimensional TLC can profile lipids, metabolites, or other small molecules from cells, tissues, and microorganisms. Comparing migration patterns helps assess sample composition, monitor purification, and identify biochemical changes associated with immune responses or pathogen activity.

Composition of polar lipid extracts and the fatty acid composition of individual glycerolipids are determined in a simple and robust lipid profiling experiment. For this purpose, glycerolipids are isolated by thin layer chromatography and subjected to transmethylation of their acyl groups. Fatty acyl methylesters are quantified by gas-liquid chromatography.
