Colorectal Tissue Isolation

Colorectal tissue isolation is the laboratory process of separating colon or rectal tissue from surrounding structures and preparing it for cellular, molecular, or histological analysis. It typically involves careful dissection followed by washing, trimming, and, when required, mechanical or enzymatic dissociation to release defined cell populations while preserving tissue architecture or cell viability; sterile handling and controlled conditions help limit contamination and degradation. Isolated colorectal tissue supports investigations of epithelial biology, inflammation, cancer, barrier function, and patient-derived models, including organoids and ex vivo assays. Standardized isolation improves sample quality and reproducibility, strengthening translational studies of gastrointestinal disease and therapeutic response.

We describe the establishment of orthotopic colorectal tumors via injection of tumor cells or organoids into the cecum of mice and the subsequent isolation of circulating tumor cells (CTCs) from this model.

Laser Microdissection: A Technique to Isolate Invading Colorectal Cancer cells from a 3D Organotypic Culture Model

2023

The video describes the laser dissection of the 3D organotypic co-culture model with micron-level precision. In 3D organotypic models, stromal constituents such as fibroblasts are 3-dimensionally co-cultured with cancer epithelial cells. The 3D model serves as an experimental tool that enables invasion and cancer-stroma interactions to be studied in near-physiological conditions.

CTC Isolation from a Whole Blood Sample: A Method to Obtain CTCs from Murine Colorectal Cancer Model

2023

This video describes a detailed protocol for isolating circulating tumor cells from the whole blood sample of a euthanized colorectal tumor mouse model. These isolated CTCs can be cultured and used for intended downstream analysis.

Two techniques can be used to establish this model: injection of a cancer cell suspension into the cecal wall or transplantation of a piece of subcutaneous tumor onto the cecum. This model is useful for studying the natural progression of colorectal cancer and testing new therapeutic agents against colorectal cancer.

Segmental Adeno-Cre Infection: A Technique to Generate Isolated Colorectal Cancer using Genetically Engineered Mouse Models

2023

This video describes the generation of a colorectal mouse model by segmental adeno-cre infection. The infected colon segment leads to tumorigenesis within this segment of the mucosa, resulting in adenomas ultimately progressing to invasive and metastatic carcinoma. This model is an attractive platform for studies of cancer biology and preclinical therapeutic trials.
