Muscularis Externa

The muscularis externa is the smooth muscle layer of the gastrointestinal tract that propels and mixes luminal contents, making it essential for digestion and normal gut function. It typically contains inner circular and outer longitudinal muscle layers, whose coordinated contractions narrow and shorten the intestinal wall to generate peristalsis and segmentation; in the stomach, an additional inner oblique layer supports mechanical mixing. Neural signals from the enteric nervous system, together with hormonal and local stimuli, regulate this activity. In medicine, the muscularis externa is important for understanding dysmotility, obstruction, achalasia, and other disorders that disrupt gastrointestinal movement.

Postoperative ileus (POI) is a complication of abdominal surgery leading to increased morbidity and a prolonged hospital stay. Because prophylactic or therapeutic strategies are lacking intensified research is necessary. Therefore we established a standardized and feasible mouse model to investigate the pathophysiology of POI and to study potential therapeutic options.

The interstitial cells of Cajal (ICC) are the pacemaker cells of the gastrointestinal (GI) tract. They form complex networks between smooth muscle cells and post-ganglionic neuronal fibers to regulate GI contractility. Here, we present immunofluorescence methods cross-sectional and whole-mount visualization of murine ICC networks.

Intracecal Tumor Cell Injection: A Technique to Deliver Cancer Cells for Establishing Orthotopic Colorectal Cancer Mouse Model

2023

The video describes the technique for intracecal injection of cancerous cells to develop an orthotopic mouse model with highly uniform tumors. Therefore, the orthotopic mouse model allows realistic simulation of metastasis, providing a valuable tool to study the phenotype changes during tumor growth and dissemination.
